Cronotol 250 is a generic disulfiram 250mg tablet designed as a pharmacological aid in the treatment of chronic alcoholism. It works by interfering with alcohol metabolism in the body, creating an acute sensitivity to ethanol. When alcohol is consumed while on disulfiram, it leads to a series of unpleasant physiological reactions—such as nausea, vomiting, palpitations, and flushing—thereby strongly deterring the individual from drinking. Unlike medications that aim to reduce cravings, disulfiram serves as an aversive agent, making alcohol consumption physically intolerable. Cronotol 250 is most effective in individuals with a sincere commitment to abstinence and is commonly used as part of a structured and supervised long-term sobriety program. This medication reinforces psychological motivation by linking alcohol consumption with immediate and adverse outcomes. When used in combination with counseling, behavioral therapy, and social support, it significantly increases the likelihood of long-term sobriety in alcohol-dependent patients.

Disulfiram exerts its therapeutic effect by irreversibly inhibiting the enzyme aldehyde dehydrogenase, which is responsible for the second step of alcohol metabolism. When alcohol is consumed, it is converted into acetaldehyde—a toxic intermediate—which accumulates rapidly due to disulfiram’s enzyme-blocking action. This accumulation leads to intense adverse effects such as sweating, nausea, chest pain, palpitations, and low blood pressure, typically occurring within 10 to 30 minutes of alcohol intake. The deterrent effect reinforces behavioral change by creating a strong negative association with alcohol. Usual Adult Dose for Alcohol Dependence: → Initial dose: 500 mg orally once daily for 1 to 2 weeks → Maintenance dose: 250 mg orally once daily (range: 125 mg to 500 mg) → Maximum dose: 500 mg once daily Duration of therapy varies depending on patient response and may extend over several months or years. The tablet is usually taken in the morning, but if sedation occurs, it can be taken in the evening. Dosage adjustments may be made based on tolerability. Important: Alcohol (including trace amounts in sauces, cough syrups, or hygiene products) must be avoided during treatment and for at least 14 days after stopping disulfiram.
