Egolix 150 Tablet contains Elagolix 150 mg, a non-peptide gonadotropin-releasing hormone (GnRH) receptor antagonist that rapidly inhibits endogenous estrogen production. It is primarily indicated for the treatment of moderate to severe endometriosis-related pain, including dysmenorrhea, chronic pelvic pain, and dyspareunia. Elagolix offers a novel, orally administered alternative to injectable GnRH agonists, allowing for a reversible, dose-dependent suppression of ovarian hormone levels. It is also effective in reducing uterine fibroid symptoms such as heavy menstrual bleeding, especially when used in high doses or in combination with hormonal add-back therapy. Its favorable pharmacokinetic profile enables individualized treatment durations tailored to patient-specific risk factors like bone density or liver function.

Elagolix competitively binds to pituitary GnRH receptors, inhibiting LH and FSH secretion. This suppression lowers circulating estradiol and progesterone levels, reducing the hormonal stimulation that drives endometrial and fibroid tissue growth. Unlike traditional GnRH agonists, Elagolix acts within hours and does not cause an initial hormone flare. At 150 mg once daily, it maintains effective symptom control while minimizing adverse effects. When used at higher dosages (e.g., 200 mg BID), deeper suppression is achieved, suitable for severe symptoms but limited to short durations due to the risk of decreased bone mineral density.
Usual Adult Dose for Endometriosis: - 150 mg orally once daily for a maximum of 24 months For Women with Dyspareunia: - 200 mg orally twice daily for up to 6 months Administration Notes: - Exclude pregnancy before starting therapy, or initiate within 7 days of menstruation onset. - Use the lowest effective dose to balance symptom control and side effect risk. - Consider co-prescription with estradiol/norethindrone acetate in long-term regimens to prevent hypoestrogenic complications such as bone loss. - For women with moderate hepatic impairment or those on high-dose therapy, limit treatment to 6 months. - This medication does not provide contraception; recommend non-hormonal methods during use.
