Donamem 10 is a fixed-dose combination of donepezil 5mg and memantine 10mg, designed for the treatment of moderate to severe Alzheimer’s disease. This dual-action therapy integrates two evidence-based mechanisms to provide comprehensive management of cognitive decline and behavioral symptoms in neurodegenerative disorders. Donepezil enhances cholinergic transmission by inhibiting acetylcholinesterase, thereby sustaining acetylcholine levels necessary for memory and learning. In parallel, memantine acts as an NMDA receptor antagonist, mitigating glutamate-induced excitotoxicity, a key factor in neuronal death. This synergistic combination supports not only memory and orientation but also reduces aggression, confusion, and loss of function. Donamem 10 is especially helpful in patients who have already shown a stable response to separate dosing of memantine and donepezil, offering a simplified regimen that improves medication adherence and long-term therapeutic outcomes.

Donamem 10 combines two pharmacologically distinct actions. Memantine selectively blocks NMDA receptors, which are overstimulated in Alzheimer’s disease due to excessive glutamate, and protects neurons from calcium-mediated toxicity. Together, they slow disease progression, enhance quality of life, and help patients retain independence in daily routines. This combination is safe for long-term use and improves functional outcomes in moderate to advanced dementia.
For patients stabilized on memantine (10 mg twice daily or 28 mg ER once daily) and donepezil 10 mg once daily: → Switch to a fixed-dose combination of memantine 28 mg ER – donepezil 10 mg, taken once daily in the evening For patients with severe renal impairment: → Start with memantine 5 mg twice daily or 14 mg ER once daily + donepezil 10 mg once daily before transitioning This combination should be initiated the day after the final separate doses of memantine and donepezil. Swallow whole with water. May be taken with or without meals. Do not split or crush the tablet. Follow all medical instructions precisely.
