Donfox-M 5/10 is a fixed-dose combination of donepezil 5mg and memantine 10mg, formulated to provide enhanced management of moderate to severe Alzheimer’s disease. By uniting the cholinergic-boosting properties of donepezil with the neuroprotective action of memantine, this medication targets the two central mechanisms of cognitive decline: acetylcholine deficiency and glutamate-induced excitotoxicity. Donepezil increases acetylcholine levels by inhibiting the enzyme acetylcholinesterase, which is crucial for memory formation and cognitive clarity. Memantine, meanwhile, blocks overactive NMDA receptors to reduce glutamate-mediated neuron damage, improving brain resilience and long-term function. Donfox-M 5/10 helps improve memory, language skills, orientation, and behavior in patients whose cognitive symptoms have progressed beyond the early stage. It also reduces episodes of confusion, apathy, and restlessness, thereby improving the quality of life and independence of patients and easing caregiver burden.

Donfox-M 5/10 combines two well-established compounds: donepezil, a cholinesterase inhibitor that boosts levels of acetylcholine in the synapses, and memantine, an NMDA receptor antagonist that regulates the excessive glutamate activity commonly seen in Alzheimer’s patients. Together, these agents not only improve neural communication and protect against further cell damage but also help restore a degree of mental clarity and emotional stability. The combination is especially effective for individuals in the intermediate to late stages of dementia and has demonstrated good safety in long-term use.
For patients stabilized on memantine (10 mg twice daily or 28 mg ER once daily) and donepezil 10 mg once daily: → Transition to memantine 28 mg ER – donepezil 10 mg combination, taken once daily in the evening In patients with severe renal impairment: → Use memantine 5 mg twice daily or 14 mg ER once daily with donepezil 10 mg before initiating combination This treatment should begin the day after stopping the separate administration of memantine and donepezil. Swallow whole with water. May be taken with or without food. Do not crush or split the tablet.
Store at temperatures below 25°C in a dry, well-ventilated area. Protect from light, heat, and moisture. Keep in the original packaging until ready to use. Store out of reach of children.
Common side effects: nausea, fatigue, dizziness, insomnia, headache, muscle cramps Less common effects: hallucinations, slow heart rate, hypertension, agitation, gastrointestinal discomfort If serious symptoms such as fainting or severe confusion occur, discontinue use and consult a healthcare provider immediately.
