Fempro 2.5 is an oral non-steroidal aromatase inhibitor containing letrozole 2.5mg, designed to reduce systemic estrogen production by blocking the aromatase enzyme. As estrogen fuels the growth of hormone receptor-positive (HR+) breast cancer, letrozole serves as a cornerstone of endocrine therapy in postmenopausal women. It is indicated for both adjuvant and metastatic settings and can be used either as monotherapy or in combination with targeted agents such as CDK4/6 inhibitors. Letrozole's long-term benefits in recurrence prevention have made it a standard of care in hormone-sensitive breast cancer. Additionally, it has gained attention in reproductive medicine for its role in inducing ovulation among women with polycystic ovary syndrome (PCOS) and other ovulatory disorders.

Letrozole selectively inhibits the aromatase enzyme, which converts androgens to estrogens in peripheral tissues. In postmenopausal women, this is the primary source of estrogen. By drastically lowering circulating estrogen levels, letrozole deprives hormone-dependent breast cancer cells of their growth signal. The drug has minimal effect on adrenal or ovarian steroidogenesis, making it safer for long-term use. It also demonstrates a favorable bone safety profile when monitored appropriately. In fertility treatment, its estrogen-lowering action indirectly stimulates the hypothalamic-pituitary-ovarian axis, promoting endogenous FSH release and ovulation.
Standard Dose in Breast Cancer: → Letrozole: 2.5 mg orally once daily → Take continuously throughout a 28-day cycle without break → Duration varies: from 5 years to 10 years depending on risk assessment and recurrence history → May be initiated after surgery (adjuvant) or prior to surgery (neoadjuvant) In Combination Therapy: → With Ribociclib: 600 mg ribociclib daily for 21 days, followed by 7 days off (28-day cycle) → Letrozole is taken daily without interruption during the same cycle → Appropriate for pre-, peri-, and postmenopausal patients when combined with ovarian suppression Ovulation Induction (Off-label use): → Typically 2.5–5 mg orally daily for 5 days, starting on day 3 or 5 of menstrual cycle → Dosage and duration may vary based on ovulatory response and clinician guidance Administration Tips: → May be taken with or without food → If a dose is missed, Take as soon as remembered unless it’s close to the next dose. Do not double up. → Use of reliable contraception is recommended during therapy if pregnancy is not intended
