Kryxana 200 is an oral anticancer therapy containing ribociclib 200mg, a selective cyclin-dependent kinase 4/6 (CDK4/6) inhibitor. It is specifically designed to halt cancer cell proliferation by interrupting the cell cycle at the G1/S transition, thereby inducing tumor cell arrest and apoptosis. This agent is primarily indicated for hormone receptor-positive (HR+), HER2-negative advanced or metastatic breast cancer and is always administered in combination with endocrine therapy such as aromatase inhibitors or fulvestrant. Its proven ability to significantly improve both overall survival (OS) and progression-free survival (PFS) has established ribociclib as a cornerstone of modern targeted breast cancer therapy. Kryxana 200 is administered on a 28-day cycle consisting of 21 days of dosing followed by 7 days off.

Ribociclib's inhibition of CDK4/6 disrupts cancer cell growth signaling and enhances the efficacy of endocrine agents. > Premenopausal Women In premenopausal women, Kryxana is co-administered with luteinizing hormone-releasing hormone (LHRH) agonists such as goserelin to induce ovarian suppression, enabling effective endocrine therapy across all age groups. - AntiCancer Drugs > Other Solid Tumors with CDK Dysregulation Investigational use includes combination regimens in malignancies like non-small cell lung cancer, pancreatic adenocarcinoma, and endometrial carcinoma where dysregulation of the CDK4/6 pathway is implicated. It remains under active clinical evaluation in these settings.
Ribociclib blocks CDK4 and CDK6, two key enzymes that regulate the progression of the cell cycle from G1 to S phase. By inhibiting this checkpoint, it prevents cellular DNA replication and tumor proliferation. Unlike traditional chemotherapy, ribociclib is a targeted agent with less systemic toxicity and is suitable for long-term oral administration. It selectively targets hormone-sensitive cancers without suppressing the entire immune system, making it an ideal component in combination endocrine therapy. The cyclic administration schedule helps to minimize cumulative toxicity while maintaining antitumor efficacy.
Early Breast Cancer (Stage II/III, High Recurrence Risk): → 400 mg orally once daily for 21 consecutive days → Followed by 7 days off (28-day treatment cycle) → Continue for up to 3 years or until recurrence or unacceptable toxicity → Must be given with an aromatase inhibitor (consult respective guidelines) Advanced/Metastatic Breast Cancer: → 600 mg orally once daily for 21 consecutive days → Followed by 7 days off (28-day treatment cycle) → Continue until disease progression or intolerable adverse events → To be combined with endocrine therapy: fulvestrant or aromatase inhibitor Important Instructions: → Take the dose at the same time each day → May be taken with or without food → If vomiting occurs after dosing, Do not retake the dose → If a dose is missed by more than 6 hours, Skip and resume next dose on schedule. Do not double up Monitoring: → Regular ECGs required for QT interval monitoring → Monitor liver function tests and neutrophil counts periodically → Dose adjustments may be needed based on toxicity profile
