Letroz 2.5 is a tablet formulation containing letrozole 2.5mg, a highly selective non-steroidal aromatase inhibitor widely used in the treatment of hormone receptor-positive (HR+) breast cancer. By targeting and inhibiting the aromatase enzyme, Letroz 2.5 significantly reduces systemic estrogen levels, effectively starving estrogen-dependent tumors of their growth stimulus. It is commonly prescribed for postmenopausal women in early-stage breast cancer as adjuvant therapy, in advanced stages as part of endocrine combination regimens, and also as a neoadjuvant option to shrink tumors before surgery. Letroz 2.5’s role has expanded beyond oncology, gaining recognition in reproductive medicine as an ovulation induction agent for women with infertility, particularly those affected by polycystic ovarian syndrome (PCOS).
- AntiCancer Drugs > HR+ Breast Cancer in Postmenopausal Women Indicated for the endocrine treatment of postmenopausal women with hormone receptor-positive breast cancer. Letrozole suppresses estrogen biosynthesis, which is essential for the growth of HR+ tumors. It is a standard in long-term adjuvant therapy and often used for 5–10 years following surgery to prevent recurrence. > Advanced or Metastatic Breast Cancer Used in combination with agents like fulvestrant, ribociclib, or palbociclib for the first-line treatment of HR+/HER2- metastatic breast cancer. Letrozole forms the endocrine backbone of these regimens, improving both progression-free and overall survival. - FEMTECH > Ovulation Induction for Infertility Letrozole is commonly used off-label in infertility management for women with ovulatory disorders. It promotes ovulation by reducing estrogen feedback on the hypothalamus, increasing FSH secretion. Compared to clomiphene, it is associated with higher monofollicular response and reduced risk of multiple pregnancy.
Letrozole acts by inhibiting the aromatase enzyme that converts androgens into estrogens in peripheral tissues. Since estrogen plays a central role in the progression of HR+ breast cancer, its suppression slows tumor growth and recurrence. In fertility treatment, the temporary drop in estrogen removes negative feedback on the hypothalamic-pituitary axis, stimulating ovulation. Letrozole is well tolerated for extended durations, has minimal impact on adrenal steroid production, and is ideal for long-term use in oncology and short-cycle use in reproductive health.
Standard Dose for Breast Cancer: → Letrozole: 2.5 mg orally once daily → Administer continuously throughout each 28-day cycle without breaks → Used for adjuvant, neoadjuvant, or metastatic therapy depending on clinical stage → Treatment may continue for 5 to 10 years based on recurrence risk Combination with Ribociclib: → Ribociclib: 600 mg orally once daily for 21 days, followed by 7 days off → Letrozole: 2.5 mg orally once daily, without interruption → Indicated for use in premenopausal (with ovarian suppression) and postmenopausal women Ovulation Induction (Off-label Use): → Typical regimen: 2.5–5 mg orally once daily for 5 consecutive days starting on day 3 or 5 of the menstrual cycle → Ovulation monitoring via ultrasound is recommended → Dosage may be adjusted in subsequent cycles based on patient response Administration Tips: → Take at the same time every day, with or without food → If a dose is missed, Take it as soon as remembered unless close to the next dose. Do not double up → Bone mineral density and cholesterol levels should be monitored during long-term therapy
