Samtica 250 is an oral anticancer medication containing abiraterone acetate, a selective inhibitor of CYP17A1. By blocking androgen biosynthesis at multiple sites, including the testes, adrenal glands, and prostate tumor tissue, it effectively suppresses disease progression in advanced prostate cancer. Samtica is used in combination with corticosteroids to minimize mineralocorticoid excess and improve tolerability.

Samtica 250 is used in combination with prednisone in patients whose prostate cancer has progressed despite medical or surgical castration. By targeting CYP17A1, it blocks androgen production from both adrenal and intratumoral sources, significantly reducing PSA levels and extending overall survival.
> Metastatic Castration-Sensitive Prostate Cancer (mCSPC)
In patients newly diagnosed with metastatic hormone-sensitive prostate cancer, abiraterone combined with androgen deprivation therapy (ADT) has demonstrated superior outcomes in delaying progression. It is increasingly adopted as a frontline option, either replacing or complementing docetaxel chemotherapy in high-risk patients.
> Biochemical Relapse or Non-Metastatic CRPC (Off-label)
Although not officially indicated, abiraterone is under investigation for early intervention in patients with rising PSA and no radiographic metastasis. This off-label use aims to delay metastasis and extend progression-free survival.
Samtica 250 inhibits androgen synthesis by selectively targeting CYP17A1. It reduces tumor burden and lowers PSA levels in mCRPC, delays disease progression in both hormone-sensitive and hormone-resistant prostate cancer, and enhances overall survival when combined with corticosteroids and ADT.
For metastatic CRPC, the usual adult dose is 1000 mg orally once daily with methylprednisolone 5 mg twice daily.
For metastatic high-risk CSPC, the dose is 1000 mg orally once daily with methylprednisolone 5 mg once daily.
Important Notes:
Take on an empty stomach (at least 1 hour before or 2 hours after meals).
Corticosteroid co-administration is required to prevent mineralocorticoid-related side effects.
Patients should continue receiving a GnRH analog or have undergone surgical castration.
Avoid substituting between different abiraterone formulations due to dosing variability.
Store below 25°C in a dry place, protected from light and moisture. Keep out of reach of children.
Common adverse effects include fatigue, joint swelling, hypertension, hypokalemia, and elevated liver enzymes. Serious risks include adrenal insufficiency, hepatotoxicity, and cardiac arrhythmia. Symptoms of mineralocorticoid excess, such as fluid retention and increased blood pressure, may occur; co-administration of corticosteroids is required to mitigate these effects.
