Samtica 500 is a high-strength oral formulation of abiraterone acetate, developed for the treatment of advanced prostate cancer. It is a selective inhibitor of CYP17, a critical enzyme involved in androgen biosynthesis. By blocking this enzyme, abiraterone suppresses androgen production not only in the testes but also in the adrenal glands and tumor tissues, providing a comprehensive hormonal blockade that is essential in prostate cancer management.

Samtica 500 is indicated for men with mCRPC who have progressed despite surgical or medical castration. When combined with low-dose methylprednisolone, it effectively suppresses androgen production, delays disease progression, and improves overall survival.
> Metastatic Castration-Sensitive Prostate Cancer (mCSPC)
This formulation is also approved for high-risk mCSPC. It is administered alongside ADT as a first-line therapy and may be given concurrently or sequentially with docetaxel chemotherapy, depending on disease aggressiveness and patient condition.
> Biochemical Relapse or Non-Metastatic CRPC
Although not yet a standard indication, Samtica 500 is under investigation in patients with rising PSA but no visible metastases. This off-label early intervention aims to slow the transition to symptomatic or metastatic disease.
Samtica 500 inhibits CYP17, reducing androgen production from the testes, adrenal glands, and tumor tissues. By targeting the hormonal drivers of prostate cancer, it suppresses tumor progression. The drug enhances the effectiveness of concurrent androgen deprivation therapy (ADT) and provides both systemic and intratumoral androgen suppression.
For metastatic CRPC, the recommended adult dose is 500 mg orally once daily (micronized formulation) in combination with methylprednisolone 4 mg orally twice daily.
For metastatic high-risk CSPC, the dose is 500 mg orally once daily with methylprednisolone 4 mg orally once daily.
Patients must continue to receive a GnRH analog or have undergone bilateral orchiectomy prior to or during therapy. Dosing schedules may vary depending on formulation type and patient-specific condition.
Store below 25°C in a dry, cool place, away from light and moisture. Keep out of reach of children. Do not use the tablets past the expiration date indicated on the packaging.
Common adverse reactions include fatigue, hypertension, and joint swelling or discomfort. Laboratory abnormalities may involve hypokalemia, liver function abnormalities, and fluid retention. The drug may also cause adrenal insufficiency or an increased risk of infection due to steroid co-administration. Regular blood tests are recommended to monitor liver enzymes and serum potassium levels throughout therapy.
