Efudex-40 5% Cream - 40g is a topical antimetabolite therapy containing 5% fluorouracil, indicated for the non-invasive treatment of various precancerous and cancerous skin lesions. It inhibits thymidylate synthase, disrupting DNA synthesis in abnormal epidermal cells, and selectively targets rapidly dividing atypical keratinocytes. The cream initiates a controlled inflammatory cascade, ultimately leading to the destruction of dysplastic tissue and regeneration of healthy skin.
- AntiCancerDrugs > Actinic Keratosis Efudex-40 5% Cream is used to treat actinic keratosis caused by long-term sun exposure. It targets precancerous cells by inhibiting DNA replication in dysplastic keratinocytes, triggering erythema, scaling, and crusting. The treatment promotes selective lesion destruction while allowing healthy skin to recover and regenerate post-inflammation. > Superficial Basal Cell Carcinoma This formulation is approved for treating superficial basal cell carcinoma in patients where surgery is not viable due to lesion multiplicity or anatomical location. It induces local cytotoxicity followed by lesion breakdown and epithelial recovery. The therapy offers an effective alternative to surgical excision in early-stage, non-invasive basal cell carcinomas. > Bowen’s Disease Efudex-40 is also used in the management of Bowen’s disease (cutaneous squamous cell carcinoma in situ). When applied topically, it eradicates intraepidermal neoplastic cells, thereby controlling lesion progression without surgical intervention. It is especially effective for lesions in cosmetically or functionally sensitive areas. > Offlabel Dermatologic Use Fluorouracil 5% is sometimes used off-label for managing skin lesions associated with viral infections (e.g., HPV), viral warts, and treatment-resistant keloids. These applications require specialist supervision and are considered experimental.

1. Actinic Keratosis - Apply 5% cream or solution twice daily to the affected areas in a thin film - Continue treatment until the lesion undergoes erythema, erosion, and crusting (typically 2–4 weeks) - Healing may require an additional 1–2 months post-treatment - Wash and dry the area before application; wait 10 minutes before applying - Avoid eyes, mucous membranes, and broken skin - Wash hands thoroughly after each application - Lesions that do not respond should be biopsied 2. Superficial Basal Cell Carcinoma - Apply 5% cream twice daily directly over lesions - Treatment duration is typically 3 to 6 weeks, extendable up to 10–12 weeks based on response - Monitor for expected inflammatory stages: redness, scaling, erosion, and healing - Ideal for patients with lesions in cosmetically sensitive or difficult-to-operate locations 3. Bowen’s Disease - Apply 5% cream twice daily over the lesion - Recommended duration: 4 to 6 weeks or as directed by a dermatologist - Appropriate for epidermis-confined neoplasms and patients who are poor surgical candidates - Post-treatment follow-up may include histological confirmation of lesion clearance
