Erivedge 150mg is an oral anticancer medication containing vismodegib, a selective Hedgehog pathway inhibitor. It is primarily used in patients with advanced or metastatic basal cell carcinoma (BCC) who are not suitable candidates for surgery or radiation therapy. By blocking the Hedgehog signaling pathway—a critical regulator of cell growth and differentiation—Erivedge limits the proliferation of cancerous cells and promotes tumor regression. It is supplied as oral capsules to be taken once daily, offering a targeted treatment for BCC patients who have limited therapeutic options.
- AntiCancer Drugs > Advanced Basal Cell Carcinoma (BCC) Erivedge is indicated for the treatment of adults with metastatic BCC or locally advanced BCC that has recurred after surgery or who are unsuitable for surgery and radiation. It is used as a first-line targeted therapy for these cases, providing a non-invasive alternative to conventional treatments, particularly in difficult-to-operate anatomical areas or recurrent tumors.
Vismodegib works by inhibiting the Hedgehog signaling pathway, specifically targeting the Smoothened (SMO) receptor. This pathway plays a central role in the pathogenesis of basal cell carcinoma. By blocking the aberrant activation of this pathway, vismodegib halts the uncontrolled cell proliferation characteristic of advanced BCC. The drug offers sustained responses and disease control in patients who have exhausted local therapies, including surgery or radiotherapy.
Usual Adult Dose for Basal Cell Carcinoma: - 150 mg orally once a day - Continue treatment until disease progression or unacceptable toxicity Comments: - If intolerable adverse reactions occur, treatment may be withheld for up to 8 weeks to allow recovery. - The efficacy of treatment durations under 8 weeks has not been established. - Capsules should be swallowed whole with or without food. Do not open or crush the capsules.
Store at room temperature between 20–25°C (68–77°F). Protect from moisture and light. Keep the bottle tightly closed. Do not refrigerate.
Common side effects include muscle spasms, hair loss (alopecia), dysgeusia (taste changes), fatigue, nausea, and weight loss. Serious risks include amenorrhea in females and embryo-fetal toxicity. Vismodegib must not be used during pregnancy due to the risk of severe birth defects. Male and female patients must adhere to strict contraception guidelines during treatment and for several months after discontinuation.
