This therapy offers a non-surgical, targeted alternative for patients with difficult-to-treat BCC.

It provides a non-invasive, molecularly targeted option for controlling progressive or aggressive BCC lesions where traditional treatments are not viable.

Benefits & Effectiveness

Sonidegib functions by inhibiting the Hedgehog (Hh) signaling pathway, specifically binding to and inactivating the Smoothened (SMO) receptor, a key transducer in the pathway. Since abnormal activation of this signaling is central to the development and progression of BCC, blocking it results in reduced tumor cell viability and a slowing of disease progression. The drug is associated with durable response rates and helps delay further tissue damage or spread in high-risk BCC cases.

Directions for Use

Usual Adult Dose for Basal Cell Carcinoma: - 200 mg orally once daily - Continue therapy until disease progression or occurrence of unacceptable toxicity Comments: - Baseline serum creatinine kinase (CK) and renal function should be assessed prior to starting therapy. - Pregnancy testing must be performed in women of childbearing potential prior to initiation. - Capsules must be swallowed whole and should not be chewed, opened, or crushed. - May be taken with or without food.

Storage

Store at room temperature between 20–25°C (68–77°F). Protect from moisture and light. Keep the bottle tightly closed and out of reach of children. Do not refrigerate.

Possible Side Effects

Common side effects include muscle spasms, fatigue, nausea, diarrhea, weight loss, and alopecia. More serious risks involve elevated creatine kinase levels, which may indicate muscle toxicity. Regular monitoring is essential to avoid rhabdomyolysis or renal impairment. Reproductive toxicity is a major concern—Odomzo is contraindicated in pregnancy and effective contraception must be used during and after treatment.
