Celdaz 200mg Injection is a cytotoxic anticancer drug containing Dacarbazine, an alkylating agent classified under triazenes. Administered via intravenous infusion, Dacarbazine acts as a prodrug, requiring hepatic activation to exert its therapeutic effect. It disrupts DNA synthesis in rapidly dividing malignant cells by methylating guanine residues, resulting in cell cycle arrest and apoptosis. This agent is particularly indicated for advanced-stage malignancies, including metastatic malignant melanoma and Hodgkin's lymphoma, and is a critical component of combination chemotherapy regimens.
AntiCancer Drugs > Metastatic Malignant Melanoma and Hodgkin's Disease
Celdaz is primarily used in the palliative treatment of metastatic malignant melanoma, helping to slow disease progression and improve symptom control. It is also a key component of combination regimens such as ABVD for relapsed or refractory Hodgkin’s lymphoma. Due to its systemic cytotoxicity, it is typically reserved for cases not responsive to front-line treatments or where other options are limited.
Key Benefits / Mechanism
Undergoes hepatic metabolism to form the active compound MTIC
Methylates DNA and inhibits RNA and protein synthesis in tumor cells
Induces cytotoxicity and cell death, especially in rapidly proliferating cancer cells
Non-cell cycle specific, allowing broad-spectrum activity across multiple tumor types
Historically effective in melanoma and Hodgkin’s lymphoma, even with the advent of targeted therapies
Usual Adult Dose for Malignant Melanoma:
2 to 4.5 mg/kg IV once daily for 10 days; repeat every 4 weeks OR
250 mg/m² IV once daily for 5 consecutive days; repeat every 3 weeks
Use: For metastatic malignant melanoma
Usual Adult Dose for Hodgkin's Disease:
150 mg/m² IV once daily for 5 days in combination chemotherapy; repeat every 4 weeks OR
375 mg/m² IV on Day 1 in combination chemotherapy; repeat every 15 days
Use: For Hodgkin’s disease as part of second-line combination chemotherapy
Comments:
Administer by slow intravenous infusion to minimize vein irritation and reduce phlebitis risk
Antiemetic premedication is strongly advised due to high emetogenic potential
Hematologic monitoring is essential due to dose-dependent bone marrow suppression
Dose adjustments may be necessary in patients with hepatic or renal dysfunction
Not recommended for use in pediatric patients unless strictly indicated
