Product Description

Pomide 1 is a generic formulation of pomalidomide (1 mg), a potent immunomodulatory drug (IMiD) used in the treatment of relapsed or refractory multiple myeloma (RRMM) and Kaposi’s sarcoma. It is specifically indicated for patients whose disease has progressed despite prior treatment with lenalidomide and proteasome inhibitors. Pomalidomide works by modulating immune responses, inhibiting angiogenesis, and exerting direct anti-myeloma effects, making it an essential option for late-line therapy in resistant hematologic malignancies.

Indications & Uses

AntiCancer Drugs > Relapsed or Refractory Multiple Myeloma
Pomide 1 is indicated for patients with multiple myeloma who have received at least two prior therapies, including lenalidomide and a proteasome inhibitor, and have shown disease progression. When used in combination with dexamethasone, it enhances treatment response and improves survival outcomes in relapsed or resistant cases.

> Triple-Class Refractory Multiple Myeloma
Pomide 1 is considered a last-resort therapy for high-risk patients who are refractory to all three major drug classes (IMiDs, PIs, and anti-CD38 antibodies). Ongoing clinical trials are evaluating its use in combination with newer monoclonal antibodies such as daratumumab and agents like carfilzomib to overcome multi-drug resistance.

> Kaposi’s Sarcoma
Pomide 1 is also used in adult patients with AIDS-related Kaposi’s sarcoma who have failed HAART therapy, as well as in HIV-negative patients. Through its anti-angiogenic and immunomodulatory activity, it helps reduce tumor burden and suppress lesion progression.

Benefits & Effectiveness

Pomalidomide is a third-generation immunomodulatory drug (IMiD) that enhances the function of T-cells and natural killer (NK) cells, suppresses inflammatory cytokines, and inhibits angiogenesis. These multi-pronged actions contribute to its strong efficacy in relapsed or refractory multiple myeloma and Kaposi’s sarcoma, even in patients resistant to previous lines of therapy. Its mechanism of action provides therapeutic benefit and offers hope to patients who are unresponsive to standard regimens.

Directions for Use

For multiple myeloma, the usual adult dose is 4 mg orally once daily on Days 1 through 21 of each 28-day cycle. It must always be co-administered with dexamethasone, and treatment should be continued until disease progression or the development of unacceptable toxicity.

For Kaposi’s sarcoma, the recommended dose is 5 mg orally once daily on Days 1 through 21 of each 28-day cycle. Therapy is continued until disease progression or unacceptable toxicity. In patients with AIDS-related KS, HAART should be maintained throughout pomalidomide therapy.

Additional Instructions:
Pomalidomide is highly teratogenic; therefore, strict contraception and full REMS compliance are mandatory. Routine monitoring of blood counts is essential due to the risk of myelosuppression. Prophylactic anticoagulation should be considered in patients at increased risk of thromboembolic events.

Storage

Store below 25°C in a tightly closed container, protected from moisture and light. Keep in original blister until use. Handle with gloves if necessary, and avoid exposure to crushed or broken capsules.

Possible Side Effects

Common side effects include neutropenia, anemia, thrombocytopenia, fatigue, constipation, and upper respiratory tract infections. Other risks include thromboembolic events, peripheral neuropathy, and rash. Routine lab monitoring is crucial to manage hematologic toxicity.

FAQs
