Product Description

Bosulif 100 is an oral tyrosine kinase inhibitor (TKI) containing bosutinib, specifically developed for the treatment of Philadelphia chromosome-positive chronic myeloid leukemia (Ph+ CML). It is especially effective in patients who are resistant or intolerant to previous TKIs such as Imatinib, Dasatinib, or Nilotinib. Bosutinib inhibits both BCR-ABL and SRC family kinases, offering an alternative therapeutic strategy in advanced or high-risk disease stages. Recent clinical data have supported its use as a frontline treatment in select newly diagnosed cases.

Indications & Uses

- AntiCancer Drugs > Chronic Myeloid Leukemia (Ph+ CML) Indicated for patients who have developed resistance or intolerance to prior tyrosine kinase inhibitor therapy. It demonstrates a significant major molecular response (MMR), including in cases with BCR-ABL mutations (except T315I). > Newly Diagnosed CML Approved in some regions as a first-line therapy based on progression-free survival (PFS) and long-term tolerability. Particularly effective against a wide range of BCR-ABL mutations. > Accelerated Phase & Blast Crisis CML Used in patients with advanced-phase or blast crisis CML to suppress disease progression and as part of multi-line salvage protocols.

Benefits & Effectiveness

Bosutinib is a dual-action TKI that targets both BCR-ABL and SRC family kinases. Its potency against most BCR-ABL mutants—excluding T315I—makes it suitable for resistant CML cases. By suppressing disease progression in accelerated or blast phases, Bosutinib helps delay transformation to more aggressive stages. It has demonstrated durable responses and a manageable safety profile, particularly in long-term therapy.

Directions for Use

Usual Adult Dose for Chronic Myelogenous Leukemia: - For newly diagnosed chronic phase (CP) Ph+ CML: 400 mg orally once daily with food Continue treatment until disease progression or intolerance - For chronic, accelerated, or blast phase Ph+ CML with resistance/intolerance: 500 mg orally once daily with food Continue until progression or unacceptable toxicity Additional Notes: - Take with food to enhance absorption and reduce gastrointestinal discomfort. - If a dose is missed by more than 12 hours, skip the missed dose and resume the regular schedule the following day. - No dose adjustment needed for mild to moderate renal impairment, but caution is advised in severe hepatic dysfunction.

Storage

Store below 30°C in a dry place, away from moisture and direct light. Keep in the original container and out of reach of children.

Possible Side Effects

Common adverse effects include diarrhea, nausea, vomiting, abdominal pain, and elevated liver enzymes. Hematologic side effects such as thrombocytopenia, anemia, and neutropenia may occur. Long-term use may lead to hepatotoxicity or cardiovascular effects, requiring periodic monitoring of liver function tests and ECG.

FAQs

Yes, in some countries it has been approved for use in newly diagnosed Ph+ CML due to its effective molecular response and long-term disease control.
Bosutinib has broader kinase inhibition and is effective against many BCR-ABL mutations. It is often used when patients fail or do not tolerate other TKIs.
Yes, always take Bosulif 100 with food to improve absorption and reduce stomach upset.
If more than 12 hours have passed, skip the missed dose and take the next dose as scheduled the following day. Do not double the dose.
