Lenangio 25 is an oral anticancer capsule containing 25 mg of lenalidomide, a potent immunomodulatory and antineoplastic agent. It plays a central role in the treatment of hematologic cancers by inducing targeted apoptosis of malignant cells and enhancing immune-mediated cytotoxicity.
Lenangio 25 acts through modulation of the cereblon E3 ubiquitin ligase complex, leading to the degradation of transcription factors critical for tumor growth and survival. This mechanism contributes to its dual direct antitumor and immune-enhancing properties.
It is essential in both frontline and maintenance settings across various hematologic malignancies, making it a cornerstone in the management of conditions such as multiple myeloma, myelodysplastic syndromes, and lymphomas.

Lenangio 25 is widely prescribed for multiple myeloma in combination with dexamethasone, forming a foundational regimen in both newly diagnosed and relapsed cases.
It is also FDA-approved for treating anemia in patients with 5q-deletion MDS, providing a targeted therapeutic approach in this subgroup.
Furthermore, Lenangio 25 serves as a key agent in the management of relapsed mantle cell lymphoma and is integrated into combination protocols for follicular lymphoma and marginal zone lymphoma, expanding its role across diverse hematologic malignancies.

1. Multiple Myeloma (MM)
In combination with dexamethasone: 25 mg orally once daily on Days 1–21 of a 28-day cycle. Continue treatment until disease progression or intolerable toxicity.
Maintenance therapy after autologous stem cell transplant: Start at 10 mg orally once daily for 28 days per cycle (for 3 cycles). If tolerated, escalate to 15 mg daily and continue as long as beneficial.
For transplant-eligible patients: mobilize stem cells within 4 cycles. Maintenance is initiated post hematologic recovery (ANC ≥1000/µL, PLT ≥75,000/µL).
2. Myelodysplastic Syndromes (MDS)
10 mg orally once daily continuously. Dose adjustment is based on hematologic response and tolerance.
Approved for transfusion-dependent anemia in low to intermediate-1 risk MDS with confirmed 5q deletion (with or without other cytogenetic abnormalities).
3. Lymphoma
Follicular Lymphoma (FL) / Marginal Zone Lymphoma (MZL): 20 mg orally once daily on Days 1–21 of each 28-day cycle (up to 12 cycles). Administer with rituximab as per protocol.
Mantle Cell Lymphoma (MCL): 25 mg orally once daily on Days 1–21 of each 28-day cycle. Continue until disease progression or unacceptable toxicity.
Store at a temperature below 25°C (77°F) in a dry, secure location. Keep capsules in their original blister to prevent exposure to moisture. Strictly avoid handling by pregnant women or those of childbearing potential unless wearing gloves, due to its teratogenic nature.
