Product Description

Dasanat 20 is a generic formulation of dasatinib, a second-generation tyrosine kinase inhibitor (TKI) with high potency against BCR-ABL and SRC-family kinases. It is indicated primarily for hematologic malignancies harboring the Philadelphia chromosome, offering a targeted mechanism to inhibit abnormal cell proliferation. Dasanat 20 exhibits rapid cytogenetic response and is frequently selected for patients resistant or intolerant to imatinib. It also plays a crucial bridging role prior to stem cell transplant in advanced leukemias.

Indications & Uses

- AntiCancer Drugs > Philadelphia chromosome-positive leukemias (CML, Ph+ ALL) Dasanat 20 is used in adult and pediatric patients with chronic myeloid leukemia (CML) and Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph+ ALL). It is effective in both frontline and salvage settings, particularly when imatinib fails. Additionally, it is employed in blast crisis phases of CML and as a central nervous system–penetrating agent in advanced ALL.

Benefits & Effectiveness

- Potent inhibition of BCR-ABL1 kinase (including mutant forms resistant to imatinib) - SRC-family kinase blockade (e.g., LYN, FYN), suppressing leukemic signaling - Ability to penetrate sanctuary sites such as the CNS - Rapid induction of hematologic and cytogenetic remission

Directions for Use

1. Adult Dosage – Leukemia - Chronic Phase CML: 100 mg orally once daily - Accelerated Phase CML / Myeloid or Lymphoid Blast Phase CML / Ph+ ALL: 140 mg orally once daily Treatment should be continued until disease progression or unacceptable toxicity. Therapeutic goals include complete hematologic response (CHR), cytogenetic remission, and long-term disease control. Dose adjustments may be required for hematologic toxicity or fluid retention. 2. Pediatric Dosage – Leukemia - Body weight 10 to <20 kg: 40 mg orally once daily - Body weight 20 to <30 kg: 60 mg orally once daily - Body weight 30 to <45 kg: 70 mg orally once daily - Body weight ≥45 kg: 100 mg orally once daily In pediatric Ph+ ALL, dasatinib should be initiated no later than Day 15 of induction chemotherapy. Treatment is continued for up to 2 years unless relapse or toxicity occurs. Weight-based reassessment is recommended every 3 months. Tablets are not suitable for children under 10 kg.

Storage

Store in a tightly closed container at temperatures below 25°C (77°F). Protect from moisture and direct sunlight. Keep out of reach of children. Do not crush or split tablets due to potential occupational exposure risk.

Possible Side Effects

- Common: fatigue, diarrhea, headache, rash, nausea - Hematologic: neutropenia, thrombocytopenia, anemia - Serious: pleural effusion, QT prolongation, pulmonary arterial hypertension - Other: myelosuppression, fluid retention, bleeding tendency

FAQs

Yes. It is approved for first-line treatment of Ph+ chronic phase CML and can be combined with chemotherapy for newly diagnosed Ph+ ALL.
Yes, dasatinib is approved for pediatric Ph+ CML and Ph+ ALL. Doses are adjusted by weight and therapy is often prolonged.
Regular monitoring of blood counts, ECG (for QT prolongation), and fluid status is necessary. Dose interruptions may be required for toxicity.
Yes. Dasatinib crosses the blood–brain barrier better than other TKIs, offering potential protection against CNS relapse in Ph+ ALL.
