Bosutris 500 is a generic formulation of bosutinib, a second-generation tyrosine kinase inhibitor (TKI) indicated for the treatment of Philadelphia chromosome-positive chronic myelogenous leukemia (Ph+ CML). This oral anticancer agent targets both BCR-ABL and SRC kinases, making it particularly effective in patients who are resistant or intolerant to prior TKIs such as imatinib, dasatinib, or nilotinib. Bosutris 500 demonstrates a high rate of major molecular response (MMR) and provides durable clinical benefit across different stages of CML.

Bosutris 500 is approved for use in patients with Ph+ CML who are resistant or intolerant to prior therapies. By inhibiting BCR-ABL tyrosine kinase activity, it effectively suppresses leukemic proliferation, even in patients with multiple treatment failures.

> Newly Diagnosed CML
Bosutris 500 has also been approved in various regions as a first-line treatment for newly diagnosed Ph+ CML. It provides strong molecular responses, enables rapid disease control, and demonstrates favorable tolerability for long-term therapy.

> Accelerated Phase & Blast Crisis
Bosutris 500 is incorporated into treatment protocols for patients whose disease has progressed to the accelerated phase or blast crisis. Its goal is to delay further progression and help stabilize hematologic parameters prior to more intensive therapies such as stem cell transplantation.

Benefits & Effectiveness

Bosutinib acts as a dual inhibitor of BCR-ABL and SRC family kinases. This dual mechanism enables it to effectively suppress leukemic cell activity, even in patients who are refractory to other TKIs. Its efficacy extends to most BCR-ABL mutations, with the exception of T315I. By targeting multiple kinase pathways, Bosutinib helps maintain long-term remission and improves progression-free survival in both frontline and salvage therapy settings.

Directions for Use

For newly diagnosed chronic phase Ph+ CML, the usual adult dose is 400 mg orally once daily with food. Treatment should continue until disease progression or the development of unacceptable toxicity.

For chronic, accelerated, or blast phase Ph+ CML after prior therapy failure, the recommended dose is 500 mg orally once daily with food. Therapy should be maintained as long as it remains clinically effective.

Additional Instructions:
Bosutris 500 should always be taken with food to minimize gastrointestinal side effects and enhance drug absorption. If more than 12 hours have passed since a missed dose, the dose should be skipped, and the next scheduled dose resumed. Patients must not double the dose to compensate for a missed one.

Storage

Store in a dry place at room temperature, below 30°C. Protect from moisture and direct sunlight. Keep the medication out of reach of children and in its original packaging.

Possible Side Effects

Common side effects include diarrhea, nausea, vomiting, stomach pain, and increased liver enzymes. Patients may also experience fatigue, rash, and hematologic abnormalities such as thrombocytopenia or neutropenia. Liver function and blood counts should be monitored regularly throughout treatment.
