Product Description

Spnib 100 is a generic formulation of dasatinib, an oral tyrosine kinase inhibitor (TKI) widely used in hematologic oncology. It is highly potent against BCR-ABL mutations, including those resistant to first-line treatments, and also exerts broader inhibition by targeting SRC family kinases.

Due to its ability to penetrate the central nervous system (CNS), Spnib 100 is effective in managing both chronic and aggressive forms of Philadelphia chromosome–positive (Ph+) leukemias, including advanced phases where disease control is challenging.

Indications & Uses

AntiCancer Drugs > Chronic Myeloid Leukemia (Ph+ CML)
Spnib 100 is used as a second-line treatment in patients who develop resistance or intolerance to imatinib, and it is also approved as a first-line option in certain high-risk patients. By inhibiting both BCR-ABL and SRC kinases, it delivers a rapid and deep molecular response.

> Philadelphia Chromosome–Positive Acute Lymphoblastic Leukemia (Ph+ ALL)
Spnib 100 is indicated for relapsed or refractory Ph+ ALL, either as monotherapy or in combination with chemotherapy. It is frequently used as a bridge to stem cell transplantation, with the added advantage of CNS penetration, providing prophylaxis against neuroinvasion.

Spnib 100 is administered when CML progresses into blast crisis, resembling an acute leukemia phase. It is known to induce a faster cytogenetic response than earlier TKIs such as imatinib, making it valuable in aggressive disease states.

Benefits & Effectiveness

Dasatinib is a second-generation tyrosine kinase inhibitor (TKI) with broad activity across BCR-ABL mutations and SRC family kinases. It is effective against leukemic cells harboring resistance to imatinib and has demonstrated efficacy in both the chronic and blast phases of leukemia. Its CNS activity provides the ability to address sanctuary sites often inaccessible to other TKIs.

Directions for Use

Adult Dose – Leukemia

  • Chronic Phase CML: 100 mg orally once daily

  • Accelerated/Blast Phase CML, Ph+ ALL: 140 mg orally once daily

Pediatric Dose – Leukemia

  • 10–<20 kg: 40 mg orally once daily

  • 20–<30 kg: 60 mg orally once daily

  • 30–<45 kg: 70 mg orally once daily

  • ≥45 kg: 100 mg orally once daily

Duration: Continue therapy until disease progression or unacceptable toxicity.

Additional Notes:

  • Dosing is weight-based in pediatric patients; reassess every 3 months or after weight changes.

  • Initiate Ph+ ALL treatment by day 15 of induction chemotherapy and continue for up to 2 years.

  • Tablets are not recommended for children weighing under 10 kg.

Storage

Store at room temperature (15–30°C), away from moisture and direct sunlight. Keep in original packaging to protect from light. Keep out of reach of children.

Possible Side Effects

Common side effects include myelosuppression (anemia, neutropenia), fluid retention (pleural effusion), diarrhea, skin rash, fatigue, and headache. Serious risks include QT prolongation, pulmonary arterial hypertension, and gastrointestinal bleeding. Regular blood count monitoring is advised.
