Lenalid 10 is an oral anticancer capsule containing 10mg of lenalidomide, a powerful immunomodulatory drug derived from thalidomide. It exerts multiple mechanisms of action including inhibition of angiogenesis, suppression of pro-inflammatory cytokines, and degradation of specific transcription factors via cereblon E3 ubiquitin ligase complex. Lenalid 10 is an essential therapeutic agent for hematologic malignancies such as multiple myeloma, myelodysplastic syndromes, and lymphomas, showing high efficacy in both induction and maintenance settings.
AntiCancerDrugs > Multiple Myeloma
Lenalid 10 is widely prescribed for the treatment of multiple myeloma, particularly in combination with dexamethasone. It enhances immune-mediated cytotoxicity against myeloma cells, inhibits their proliferation, and induces apoptosis. The drug is used as frontline therapy, in relapsed settings, and as maintenance after stem cell transplantation to extend remission.
Myelodysplastic Syndrome (MDS)
For patients with low to intermediate-1 risk MDS with 5q deletion, Lenalid 10 significantly reduces transfusion dependency and improves hematopoiesis. Its targeted mechanism supports normalization of bone marrow function and long-term cytogenetic response.
Lymphoma
Lenalid 10 is indicated for mantle cell lymphoma (MCL) in patients with prior therapies and is also used off-label in follicular lymphoma (FL) and marginal zone lymphoma (MZL). Combined with rituximab, it enhances response durability and improves disease control in indolent lymphoid malignancies.
Off-label Use in CLL and Hematologic Cancers
Lenalidomide has shown potential in clinical research for chronic lymphocytic leukemia (CLL) and other lymphoproliferative diseases, particularly where conventional regimens fail or are contraindicated. Its immunostimulatory and anti-angiogenic effects make it a viable adjunct in refractory cases.
Multiple Myeloma (MM)
With dexamethasone: 25 mg orally once daily on Days 1–21 of a 28-day cycle
Maintenance post-auto-HSCT: Start with 10 mg orally once daily on Days 1–28 for 3 cycles
Increase to 15 mg daily if tolerated, continue until progression or toxicity
Duration: Continue until disease progression or unacceptable adverse events
Stem cell collection: Begin within 4 cycles if planned
Myelodysplastic Syndromes (MDS)
10 mg orally once daily
Adjust dosage depending on blood counts and patient tolerance
Indicated for 5q deletion with or without additional cytogenetic abnormalities
Response assessed by transfusion needs and marrow cytogenetics
Lymphomas
Follicular Lymphoma (FL) / Marginal Zone Lymphoma (MZL): 20 mg orally once daily on Days 1–21 of a 28-day cycle (up to 12 cycles)
Co-administered with rituximab
Mantle Cell Lymphoma (MCL): 25 mg orally once daily on Days 1–21 of a 28-day cycle
Continue until disease progression or unacceptable toxicity
