Lenangio 10 is an oral anticancer capsule formulated with 10mg of lenalidomide, a potent second-generation immunomodulatory drug derived from thalidomide. It exerts multiple therapeutic actions including direct anti-proliferative effects, inhibition of tumor angiogenesis, immune system modulation, and induction of apoptosis in malignant cells. Lenangio 10 is particularly effective in treating hematologic malignancies such as multiple myeloma, myelodysplastic syndromes, and various lymphomas, playing a critical role in both initial therapy and long-term disease control.

Indications & Uses

  • AntiCancerDrugs > Multiple Myeloma
    Lenangio 10 is widely utilized in the frontline and relapsed treatment of multiple myeloma, often in combination with dexamethasone or proteasome inhibitors like bortezomib. It enhances T-cell and NK-cell cytotoxicity, inhibits cytokine production, and disrupts microenvironmental support of myeloma cells. It is also used in maintenance therapy following autologous stem cell transplant to prolong remission and improve overall survival.

  • Myelodysplastic Syndrome (MDS)
    In patients with low or intermediate-1 risk MDS with 5q deletion, Lenangio 10 has been shown to significantly reduce the need for red blood cell transfusions and improve bone marrow function. It is the first FDA-approved agent targeting this specific cytogenetic abnormality, making it a critical treatment option for transfusion-dependent anemia.

  • Lymphoma
    Lenangio 10 is indicated in relapsed or refractory mantle cell lymphoma (MCL), especially in patients who have failed prior treatments such as bortezomib. It is also used off-label in follicular lymphoma (FL) and marginal zone lymphoma (MZL), typically in combination with rituximab, offering an alternative therapeutic approach in indolent B-cell malignancies.

  • Off-label in CLL and Other Lymphoproliferative Disorders
    Emerging studies suggest the utility of lenalidomide in chronic lymphocytic leukemia (CLL) and other hematologic cancers due to its ability to modulate immune responses and disrupt leukemic cell survival pathways.

Directions for Use

  • Multiple Myeloma (MM)

    In combination with dexamethasone: 25 mg orally once daily on Days 1–21 of a 28-day cycle
    Continue until disease progression or unacceptable toxicity
    Maintenance therapy post-autologous HSCT: Start with 10 mg once daily on Days 1–28 of 28-day cycles for 3 cycles

    If tolerated, increase to 15 mg once daily and continue long-term

    Notes: For non-transplant candidates, lenalidomide is continued until disease progression

    For transplant candidates, stem cell mobilization is recommended within 4 cycles
    Blood counts must meet recovery thresholds before initiating maintenance
     
  • Myelodysplastic Syndromes (MDS)
    10 mg orally once daily
    Dosage may be adjusted based on hematologic and cytogenetic response
    Designed for use in patients with transfusion-dependent anemia and 5q deletion
  • Lymphoma
    Follicular Lymphoma (FL) / Marginal Zone Lymphoma (MZL): 20 mg orally once daily on Days 1–21 of a 28-day cycle for up to 12 cycles

    Use in combination with rituximab
    Mantle Cell Lymphoma (MCL): 25 mg orally once daily on Days 1–21 of a 28-day cycle
    Continue until progression or unacceptable toxicity

Storage

Store at room temperature below 25°C. Keep the capsules in their original blister packaging to protect from light and moisture. Ensure safe storage away from children and pregnant women due to high teratogenic risk. Dispose of unused medication in accordance with local pharmaceutical waste protocols.
