Lenalid 25 is an oral anticancer medication containing 25 mg of lenalidomide, an immunomodulatory agent primarily indicated for the treatment of hematologic malignancies.
Its mechanism involves targeting and degrading specific cellular proteins through the cereblon E3 ubiquitin ligase complex, which induces apoptosis of malignant cells. At the same time, it enhances the activity of T-cells and natural killer (NK) cells, contributing to its combined immunomodulatory and antitumor effects.
Lenalid 25 is widely used in combination therapies, serving as a cornerstone in multiple modern treatment regimens for blood cancers.

Lenalid 25 is widely used for patients with newly diagnosed or relapsed/refractory multiple myeloma, most commonly in combination with dexamethasone or bortezomib.
It is also the first FDA-approved therapy for transfusion-dependent anemia in MDS patients with 5q deletion, establishing a targeted treatment pathway.
In relapsed mantle cell lymphoma as well as follicular and marginal zone lymphomas, Lenalid 25 provides a targeted option, either as monotherapy or in combination with rituximab, expanding its role across diverse hematologic cancers.

1. Multiple Myeloma (MM)
In combination with dexamethasone: 25 mg orally once daily on Days 1–21 of each 28-day cycle. Continue until disease progression or unacceptable toxicity.
Maintenance after autologous HSCT: Begin at 10 mg once daily (Days 1–28) for 3 cycles. If well tolerated, escalate to 15 mg once daily and maintain until progression.
Stem cell mobilization should occur within the first 4 cycles in transplant-eligible patients.
Maintenance should start only after recovery of neutrophils (ANC ≥1000/µL) and platelets (PLT ≥75,000/µL).
2. Myelodysplastic Syndrome (MDS)
10 mg orally once daily, adjusted based on hematologic response and tolerance.
Indicated for transfusion-dependent anemia in low or intermediate-1 risk MDS with isolated 5q deletion, with or without additional cytogenetic abnormalities.
3. Lymphoma
Follicular Lymphoma / Marginal Zone Lymphoma (FL/MZL): 20 mg orally once daily on Days 1–21 of each 28-day cycle, for up to 12 cycles. Always used in combination with rituximab.
Mantle Cell Lymphoma (MCL): 25 mg orally once daily on Days 1–21 of each 28-day cycle. Continue until disease progression or unacceptable toxicity.
Store below 25°C (77°F) in a dry place away from direct sunlight. Keep the capsules in the original packaging to protect from moisture and avoid exposure to children or pregnant individuals due to teratogenic risk.
