Bosulif 500 is an original brand formulation of bosutinib, a second-generation oral tyrosine kinase inhibitor (TKI) specifically designed to target Philadelphia chromosome-positive chronic myelogenous leukemia (Ph+ CML). It effectively inhibits both BCR-ABL and SRC family kinases, making it a powerful option for patients with resistance or intolerance to first-line treatments. With proven long-term safety and efficacy, Bosulif 500 provides a high molecular response rate and deep remission in chronic, accelerated, and blast phases of CML.
- AntiCancer Drugs > Chronic Myeloid Leukemia (Ph+ CML) Used in patients who have failed or cannot tolerate imatinib, dasatinib, or nilotinib. Bosulif 500 suppresses leukemic cell growth by dual inhibition of BCR-ABL and SRC kinases and is approved for use in second- or third-line treatment. > Newly Diagnosed CML Bosutinib has gained approval in several regions as a first-line treatment for newly diagnosed CML due to its rapid molecular response and favorable safety profile. It demonstrates efficacy against most BCR-ABL mutations, except T315I. > Accelerated Phase & Blast Crisis In patients with advanced CML stages, Bosulif 500 is utilized to delay disease progression and achieve temporary control before hematopoietic stem cell transplantation or other escalation therapies.
Bosutinib selectively inhibits the BCR-ABL tyrosine kinase as well as SRC kinases, effectively halting the proliferation of leukemic cells in patients with Ph+ CML. It offers a high rate of major molecular response (MMR), cytogenetic remission, and long-term disease control. Especially effective in cases of prior TKI failure, it is a key agent in modern CML treatment algorithms.
Usual Adult Dose for Chronic Myelogenous Leukemia: - Newly diagnosed chronic phase Ph+ CML: 400 mg orally once daily with food Treatment should be continued until disease progression or unacceptable toxicity - Chronic, accelerated, or blast phase Ph+ CML with resistance or intolerance to prior therapy: 500 mg orally once daily with food Continue therapy as long as it remains clinically beneficial Additional Instructions: - Always take with food to enhance drug absorption and reduce gastrointestinal side effects. - If a dose is missed and more than 12 hours have passed, skip the missed dose and resume normal dosing the next day. Do not double the dose.
Store in a cool, dry place below 30°C. Keep in the original packaging to protect from light and moisture. Ensure the container is tightly closed and kept out of reach of children.
Common adverse effects include diarrhea, nausea, vomiting, and abdominal pain. Patients may also experience liver enzyme elevations, low blood counts (neutropenia, thrombocytopenia), and fatigue. Less commonly, rash, fluid retention, and headache may occur. Regular monitoring of blood counts and liver function is essential during treatment.
