Lenangio 5 is a generic oral anticancer medication containing lenalidomide 5mg, used in the management of multiple hematological malignancies. As an immunomodulatory agent, it modulates the cereblon E3 ubiquitin ligase complex, promoting apoptosis of malignant cells and enhancing immune cell-mediated tumor suppression. Lenangio 5 plays a crucial role in frontline therapy, maintenance regimens, and relapsed settings across diseases such as multiple myeloma, MDS, and certain lymphomas.
AntiCancerDrugs > Multiple Myeloma
Lenangio 5 is indicated for the treatment of multiple myeloma, particularly in combination with dexamethasone. It is used in both newly diagnosed and relapsed/refractory cases. The drug induces cytotoxic effects via apoptosis and inhibits tumor-supporting cytokines. For patients undergoing autologous stem cell transplantation, it is also prescribed as a maintenance therapy to prolong remission duration and improve survival outcomes.
Myelodysplastic Syndrome (MDS)
Lenangio 5 is approved for patients with transfusion-dependent anemia associated with low- or intermediate-1-risk MDS with chromosome 5q deletion. It improves red blood cell production, reduces transfusion requirements, and aids marrow recovery by modifying the tumor microenvironment and cellular differentiation pathways.
Mantle Cell Lymphoma (MCL)
In patients who have undergone at least two prior therapies, including bortezomib, Lenangio 5 is used as a second-line or beyond treatment. It acts by suppressing neoplastic cell proliferation and enhancing T-cell activity, often delaying disease progression in refractory cases.
Off-label Use in Other Lymphomas
Lenangio 5 may be used off-label in conditions such as chronic lymphocytic leukemia (CLL), follicular lymphoma (FL), and marginal zone lymphoma (MZL), usually in combination with rituximab. These regimens leverage lenalidomide’s immune-enhancing properties to improve response in indolent lymphoid cancers.
Multiple Myeloma
In combination with dexamethasone: 25 mg orally on Days 1–21 of a 28-day cycle
Maintenance post-autologous stem cell transplant: Start with 10 mg daily on Days 1–28 for 3 cycles; may increase to 15 mg daily based on tolerance
Continue until disease progression or toxicity
Stem cell mobilization should occur within 4 cycles; initiate maintenance once ANC ≥1000/µL and PLT ≥75,000/µL
Myelodysplastic Syndromes (MDS)
10 mg orally once daily
Adjust dosage based on clinical and hematologic response
Use: For anemia in 5q-deletion MDS (± other chromosomal abnormalities)
Lymphomas
Follicular Lymphoma (FL) / Marginal Zone Lymphoma (MZL): 20 mg orally on Days 1–21 of each 28-day cycle, up to 12 cycles (with rituximab)
Mantle Cell Lymphoma (MCL): 25 mg orally on Days 1–21 of 28-day cycles
Continue until unacceptable toxicity or disease progression
Store below 25°C in a dry, light-protected place. Keep capsules sealed in original packaging until use. Ensure child-safe storage and avoid exposure to moisture. Do not use if the packaging is damaged or expired.
