Lenalid 5 is an oral anticancer medication containing lenalidomide 5mg, belonging to the class of immunomodulatory drugs. It is widely used in the treatment of various hematologic malignancies, particularly multiple myeloma, myelodysplastic syndromes (MDS), and relapsed or refractory lymphomas. By targeting the cereblon E3 ubiquitin ligase complex, it induces degradation of key transcription factors and enhances immune-mediated cytotoxicity. Lenalid 5 plays a pivotal role in both frontline and maintenance therapy settings due to its dual anti-tumor and immune-enhancing properties.

Indications & Uses

  • AntiCancerDrugs > Multiple Myeloma
    Lenalid 5 is used in combination with dexamethasone for treating newly diagnosed and relapsed multiple myeloma. It induces apoptosis of malignant plasma cells, inhibits angiogenesis, and stimulates T-cell and natural killer cell activity. For transplant-ineligible patients, it serves as continuous therapy, and for those undergoing autologous stem cell transplantation, it is a core maintenance option to prolong progression-free survival.

  • Myelodysplastic Syndrome (MDS)
    Lenalid 5 is approved for the treatment of transfusion-dependent anemia in patients with low- or intermediate-1-risk MDS associated with a deletion of chromosome 5q. It reduces the need for red blood cell transfusions, improves marrow function, and supports hematologic recovery through modulation of erythropoiesis.

  • Mantle Cell Lymphoma (MCL)
    Lenalid 5 is indicated for patients with MCL who have relapsed or failed at least two prior treatments, including bortezomib. It offers clinical benefit through its anti-proliferative and immunologic mechanisms, especially in patients not eligible for aggressive therapies.

  • Off-label Lymphoid Malignancies
    The drug is occasionally used off-label in chronic lymphocytic leukemia (CLL), follicular lymphoma (FL), and marginal zone lymphoma (MZL), often in combination with rituximab, where it shows enhanced immune synergy and potential clinical response in slow-growing lymphomas.

    Dose: 10 mg orally once daily
    Continue or adjust based on clinical response and blood counts
    Use: For transfusion-dependent anemia in 5q deletion MDS (with or without other cytogenetic abnormalities)

  • Lymphomas
    Follicular Lymphoma (FL) / Marginal Zone Lymphoma (MZL): 20 mg once daily on Days 1–21 of 28-day cycles, up to 12 cycles (with rituximab)
    Mantle Cell Lymphoma (MCL): 25 mg once daily on Days 1–21 of each 28-day cycle
    Continue until disease progression or intolerable side effects

Storage

Store below 25°C in a dry, cool environment away from direct sunlight. Keep capsules in original blister packaging. Ensure child-resistant storage. Do not use if seal is broken or expired.
