Bosulif 400 is an oral tyrosine kinase inhibitor (TKI) designed for the treatment of Philadelphia chromosome-positive chronic myeloid leukemia (Ph+ CML). It belongs to the class of second-generation TKIs and specifically targets BCR-ABL fusion proteins as well as SRC family kinases. This dual inhibition mechanism allows for effective disease control in patients resistant or intolerant to first-line therapies like Imatinib, Dasatinib, or Nilotinib. Bosulif 400 is also recognized as a first-line option in some countries, with clinical data supporting its efficacy and safety in newly diagnosed patients.

Bosutinib delivers its anticancer activity by binding to and inhibiting the BCR-ABL tyrosine kinase, which drives the pathogenesis of Ph+ CML. Its added effect on SRC family kinases makes it beneficial in more refractory cases. Clinical studies have demonstrated a high Major Molecular Response (MMR) and sustained disease suppression, even in later disease stages or those with prior TKI failure.
Usual Adult Dose for Chronic Myelogenous Leukemia: - For newly diagnosed chronic phase Ph+ CML: 400 mg orally once daily with food Treatment should be continued until disease progression or intolerance - For chronic phase, accelerated phase, or blast phase Ph+ CML with prior TKI resistance/intolerance: 500 mg orally once daily with food Continue until clinical progression or unacceptable toxicity Additional Instructions: - Always take Bosulif with food to enhance absorption and minimize gastrointestinal side effects. - If a dose is missed and more than 12 hours have passed, skip the missed dose and take the next dose as scheduled. Do not double up.
Store below 30°C in a dry, dark place. Keep the tablets in their original packaging until use. Avoid exposure to excessive heat or moisture, and keep out of reach of children.
Common side effects include diarrhea, nausea, vomiting, abdominal pain, and fatigue. Liver enzyme elevation, neutropenia, and thrombocytopenia may also occur. Regular monitoring of blood counts and liver function is essential during therapy. Rash and fluid retention are less common but possible.
