Product Description

Spnib 20 is a generic formulation of dasatinib used in the treatment of Philadelphia chromosome–positive (Ph+) leukemias, including chronic myeloid leukemia (CML) and acute lymphoblastic leukemia (Ph+ ALL). As a potent second-generation tyrosine kinase inhibitor (TKI), it exerts strong inhibitory effects on both BCR-ABL and SRC-family kinases, providing rapid cytogenetic responses and effective CNS penetration.

Spnib 20 is applied in both adult and pediatric settings, particularly in patients who are resistant or intolerant to prior imatinib-based therapies.

Indications & Uses

AntiCancer Drugs > Chronic Myeloid Leukemia (CML)
Spnib 20 is indicated for adult patients with chronic phase CML, including those who are newly diagnosed or who were previously treated with imatinib and developed resistance or intolerance. Its rapid onset and broader kinase coverage make it suitable as both a frontline and second-line option.

> Philadelphia Chromosome–Positive Acute Lymphoblastic Leukemia (Ph+ ALL)
In Ph+ ALL, Spnib 20 is administered as part of a combination regimen with chemotherapy, frequently serving as a bridge to hematopoietic stem cell transplantation (HSCT). Its ability to cross the blood–brain barrier adds value in reducing the risk of CNS relapse.

> Blast Crisis and Rare Leukemias
Spnib 20 is also used in patients whose CML has progressed to myeloid or lymphoid blast crisis, offering deeper and faster cytogenetic responses compared to first-generation TKIs.

Directions for Use

1. Adult Dosage – Leukemia

  • Chronic Phase CML: 100 mg orally once daily

  • Accelerated Phase CML / Blast Phase CML / Ph+ ALL: 140 mg orally once daily

Treatment should be continued until disease progression or development of unacceptable toxicity. Patients must be monitored with regular blood counts, liver enzyme tests, and ECGs. Dose adjustments may be necessary in cases of pleural effusion, hematologic toxicity, or QT prolongation.

2. Pediatric Dosage – Leukemia

  • 10–<20 kg: 40 mg orally once daily

  • 20–<30 kg: 60 mg orally once daily

  • 30–<45 kg: 70 mg orally once daily

  • ≥45 kg: 100 mg orally once daily

For Ph+ ALL, dasatinib should be initiated within 15 days of starting induction chemotherapy and continued for up to 2 years. Pediatric dosing is strictly weight-based and should be reassessed regularly. Tablets are not recommended for children under 10 kg.

Storage

Store at controlled room temperature (15°C–30°C or 59°F–86°F). Keep away from moisture and direct sunlight. Do not crush or split the tablets. Keep out of reach of children. Use proper precautions when handling tablets due to potential exposure risk.
