Dasatib 100 is a high-dose formulation of dasatinib, designed primarily for adult patients requiring full therapeutic suppression of BCR-ABL activity. As a second-generation tyrosine kinase inhibitor (TKI), it is indicated for the treatment of Philadelphia chromosome–positive (Ph+) leukemias, including chronic myeloid leukemia (CML) and acute lymphoblastic leukemia (Ph+ ALL).
Its mechanism of action includes dual inhibition of BCR-ABL and SRC family kinases, enabling broader and faster suppression of leukemic proliferation, including cases resistant to imatinib.
Dasatib 100 is also valuable in the management of advanced disease stages and in patients with CNS-involved leukemias, where penetration of sanctuary sites is critical for disease control.
AntiCancer Drugs > Chronic Myeloid Leukemia (CML)
Dasatib 100 is used as a first-line agent in high-risk or newly diagnosed Ph+ chronic phase CML, and as a second-line treatment for patients who are resistant or intolerant to imatinib. Its capacity to induce rapid cytogenetic and molecular responses makes it a preferred choice in cases requiring swift disease control or for stem cell transplant preparation.
> Philadelphia Chromosome–Positive Acute Lymphoblastic Leukemia (Ph+ ALL)
Dasatib 100, when used either in combination with chemotherapy or as monotherapy, is effective in relapsed or refractory Ph+ ALL. Its ability to cross the blood–brain barrier provides an advantage in managing CNS leukemia, and it is frequently employed as a bridge therapy prior to allogeneic hematopoietic stem cell transplantation.
> Myeloid & Lymphoid Blast Crisis
In CML patients whose disease has progressed to blast crisis, Dasatib 100 is administered to achieve cytogenetic remission and improve candidacy for curative interventions such as stem cell transplantation.
Potent BCR-ABL inhibition with broader kinase coverage
Effective against most imatinib-resistant mutations
Penetrates CNS for improved Ph+ ALL control
High-dose 100 mg tablet ensures compliance in adult patients
Clinically proven response in both chronic and advanced phases
1. Adult Dose – Leukemia
Chronic Phase CML: 100 mg orally once daily
Accelerated Phase CML / Blast Phase CML / Ph+ ALL: 140 mg orally once daily
Duration: Continue therapy until disease progression or unacceptable toxicity. Regular monitoring of blood counts, liver function, pleural effusion, and cardiovascular status is essential. Dose modification may be required based on tolerance and response.
2. Pediatric Dose – Leukemia
10–<20 kg: 40 mg orally once daily
20–<30 kg: 60 mg orally once daily
30–<45 kg: 70 mg orally once daily
≥45 kg: 100 mg orally once daily
Notes: Pediatric use follows weight-based protocols, reviewed every 3 months. In Ph+ ALL, dasatinib should be initiated by day 15 of induction chemotherapy and maintained for up to 2 years. Tablets are not recommended for patients under 10 kg due to formulation limits.
Store at 15–30°C (59–86°F) in a dry, light-protected container. Avoid exposure to humidity and heat. Keep tablets in the original blister packaging and out of reach of children. Do not crush or break tablets unless necessary for dosing adjustments under supervision.
- Hematologic: neutropenia, anemia, thrombocytopenia - Respiratory: pleural effusion, dyspnea, pulmonary hypertension - Cardiovascular: QT prolongation, peripheral edema - Gastrointestinal: diarrhea, nausea, abdominal pain - CNS: headache, dizziness - Others: skin rash, fatigue, elevated liver enzymes
