Indications & Uses

- AntiCancer Drugs > Non-Small Cell Lung Cancer (NSCLC) Used in combination with Binimetinib for the treatment of advanced or metastatic NSCLC with confirmed BRAF V600E mutation. Offers targeted suppression of tumor growth pathways and is reserved for patients who are ineligible for curative surgery. > Unresectable or Metastatic Melanoma Indicated with Binimetinib for patients with unresectable or metastatic melanoma harboring BRAF V600E or V600K mutations. Helps delay disease progression and improve overall response when first-line therapies are no longer effective. > Metastatic Colorectal Cancer (mCRC) Used with Cetuximab in patients with metastatic colorectal cancer confirmed to have BRAF V600E mutation. Provides targeted intervention in combination therapy to reduce tumor progression when standard therapies fail.

Directions for Use

Melanoma or NSCLC: - 450 mg orally once daily (usually 6 tablets) - Administered in combination with Binimetinib - Treatment continued until disease progression or unacceptable toxicity - If Binimetinib is stopped, reduce dose to 300 mg daily Colorectal Cancer (mCRC): - 300 mg orally once daily - Used in combination with Cetuximab - Treatment continued until progression or toxicity - Discontinue Encorafenib if Cetuximab is stopped General Comments: - Ensure BRAF V600E or V600K mutation confirmation via FDA-approved test before initiation - Swallow capsules whole, with or without food - Dose adjustments may be required based on tolerability and combination agent status

Storage

Store at room temperature (20°C to 25°C). Protect from moisture and heat. Keep the bottle tightly closed and out of reach of children.

Possible Side Effects

Common adverse effects include fatigue, nausea, joint pain, and rash. Serious side effects may involve QT prolongation, liver enzyme elevation, hemorrhage, and photosensitivity. Monitor ECG and liver function regularly. Risk of new primary malignancies (e.g., cutaneous squamous cell carcinoma) also exists; perform dermatologic evaluations periodically.
