Amlodipine Besilate is widely used as a first-line treatment for hypertension. It works by blocking calcium channels in blood vessel walls, resulting in vasodilation and a reduction in blood pressure. This helps prevent the risk of stroke, heart attack, and kidney damage due to high blood pressure.
Amlodipine Besilate works by inhibiting the influx of calcium into smooth muscle cells of blood vessels, leading to relaxation and dilation of the blood vessels. This results in a significant reduction in blood pressure and a decrease in the heart's workload. Additionally, it has been shown to improve blood flow in patients with coronary artery disease and reduce chest pain in angina pectoris.
Usual Adult Dose for Hypertension
→ Initial dose: 5 mg orally once a day
→ Maintenance dose: 5–10 mg orally once a day
→ Maximum dose: 10 mg/day
→ In smaller or more fragile patients, the starting dose may be 2.5 mg orally once a day
Usual Adult Dose for Angina Pectoris
→ Maintenance dose: 5–10 mg orally once a day
→ Maximum dose: 10 mg/day
Usual Adult Dose for Coronary Artery Disease
→ Maintenance dose: 5–10 mg orally once a day
→ Maximum dose: 10 mg/day
Usual Geriatric Dose for Hypertension
→ Initial dose: 2.5 mg orally once a day
→ Maintenance dose: 2.5–10 mg orally once a day
→ Maximum dose: 10 mg/day
Usual Geriatric Dose for Angina Pectoris
→ Initial dose: 5 mg orally once a day

Pediatric Dose for Hypertension (6–17 years)
→ Maintenance dose: 2.5–5 mg orally once a day
→ Maximum dose: 5 mg/day
→ Doses higher than 5 mg have not been studied in pediatric patients
→ Titration should occur over 7–14 days, adjusting according to the patient's response
Common side effects
→ Swelling in the legs or ankles
→ Dizziness
→ Headache
→ Flushing
→ Palpitations (rapid heart rate)
Rare but serious side effects
→ Low blood pressure
→ Reflex tachycardia
